Antibodies directed against the calcium-binding proteins, parvalbumin and calbindin, can be used to label distinct neuronal subgroups in the primate visual pathway. We analyzed parvalbumin immunoreactivity (P-IR) and calbindin immunoreactivity (C-IR) in the lateral geniculate nucleus (LGN) and visual cortex of the marmoset, Callithrix jacchus. We compared marmosets which were identified as having dichromatic or trichromatic color vision. Within the LGN, the density of P-IR neurones is highest in the parvocellular and magnocellular laminae, but C-IR neurones are found mainly in the koniocellular division of the LGN, that is, the interlaminar zones and S laminae. Not all interlaminar zone cells are C-IR. In the visual cortex, P-IR neurones are present in all laminae except lamina 1, in areas V1 and V2. Neurones which are strongly C-IR are mainly located in laminae 2 and 3 in V1 and V2. Lightly C-IR neurones are concentrated in lamina 4, and are more numerous in V1 than in V2. Quantitative analysis showed no differences in the density or distribution of IR neurones in either LGN or visual cortex when dichromat and trichromat animals were compared. We conclude that this functional difference is not associated with differences in the neurochemistry of calcium-binding proteins in the primary visual pathways.

We measured the variability in latency of the first spike seen in cells of the cat's lateral geniculate nucleus following the onset of a visual stimulus. We found that, in each of the 11 cells tested, this variability was significantly lower during burst than during tonic firing. We suggest that this difference confers an advantage in signal detection during burst compared to tonic firing. This complements other reported advantages of burst firing for signal detection seen in signal-to-noise ratios and in the ability to efficiently drive postsynaptic cells.

We characterized the chromatic and temporal properties of a sample of 177 red–green parvocellular neurons in the LGN of Macaca nemestrina, using large-field stimuli modulated along different directions through a white point in color space. We examined differences among the properties of the four subclasses of red–green P-cells (on- and off-center, red and green center). The responses of off-center cells lag the stimulus more than do those of on-center cells. At low temporal frequencies, this causes the phase difference between responses of the two kinds of cells to be considerably less than 180 deg. For isoluminant modulations the phases of on- and off-responses were more nearly 180 deg apart. A cell's temporal characteristics did not depend on the class of cone driving its center. Red center and green center cells have characteristically different chromatic properties, expressed either as preferred elevations in color space, or as weights with which cells combine inputs from L- and M-cones. Red center cells are relatively more responsive to achromatic modulation, and attach relatively more weight to input from the cones driving the center. Off-center cells also attach relatively more weight than do on-center cells to input from the class of cone driving the center.

The appearance of cGMP-gated cation channel protein in the postnatal rat retina has been studied by fluorescence immunocytochemistry of radial retinal sections and immunoblots of retinal membrane proteins. Channel immunoreactivity was first detectable with RCNGC1-7H2 monoclonal antibody at postnatal day 7 (PN7) by both methods. Immunocytochemical label in retinal sections was localized to the outer segments, and immunoreactivity increased with increasing age. We also compared the developmental appearance of the cGMP-gated cation channel to that of other phototransduction proteins and developmental markers. RET-P2, a monoclonal antibody recognizing the 39-kDa rds/peripherin disc protein, first labeled outer segments at PN7, coincident with cGMP-gated cation channel expression. Double labeling of the same section of PN7 rat retina with RET-P2 and R309 (a polyclonal antiserum against the rod cGMP-gated cation channel) revealed identical patterns of labelling. Similarly, double labeling with RCNGC1-7H2 and an antibody against the rod cGMP-phosphodiesterase gave coincident labeling, suggesting coordinate expression mechanisms of phototransduction proteins with each other and with outer segment structural proteins.

Direction selectivity and orientation selectivity were examined in the direction-selective (DS) and orientation- selective (OS) ganglion cells in the rabbit retina. Spike activities were recorded in vivo using tungsten-in-glass electrodes. Three types of visual stimuli (flashing slit, moving edges behind a slit, and whole-field drifting gratings) were used to distinguish these attributes. Fourier analysis was performed on data obtained using drifting gratings as proposed by Worgotter and Eysel (1987). Results from both angular and Fourier (polar angle frequency) domains were consistent. DS cells had strong directional components and varying strength of orientational components; they also had intact inhibitory surrounds. The phase of the biased orientation did not have a consistent relationship with the preferred direction. OS cells had predominant orientational components and very weak directional components. We conclude that the orientation bias does not contribute to the generation of direction selectivity, and that the mechanism of this bias of DS cells is rather different from the mechanism that generates orientation selectivity in rabbit OS cells. There are indications that it is similar to the mechanism shown to cause orientation bias in cat concentric cells.

Development and growth of V1 begins during embryogenesis and continues postnatally. The growth of V1 has direct implications on the organization of features such as the retinotopic map and the pattern of visual cortical columns. We have examined the postnatal growth and two-dimensional shape of V1 in macaque monkeys, cats, and rats. The perimeter, area, and anterior–posterior length of V1 were measured from unfolded and flattened sections from neonatal and adult animals from each of these species. Although there were substantial differences in the overall amount of postnatal growth, from 18% in macaque monkeys to more than 100% in cats, in all three species the shape of V1 did not change during development. Thus, growth of the mammalian visual cortex is well described as an isotropic expansion, so the layout of the global features, such as the arrangement of ocular dominance columns and the retinotopic map, does not need to change during development. Furthermore, quantification of the shape confirms the observations that there is a similar, egg-like oval shape to the visual cortex of these mammalian species.

Previous studies of cat visual cortex have shown that the spatiotemporal (S-T) structure of simple cell receptive fields correlates with direction selectivity. However, great heterogeneity exists in the relationship and this has implications for models. Here we report a laminar basis for some of the heterogeneity. S-T structure and direction selectivity were measured in 101 cells using stationary counterphasing and drifting gratings, respectively. Two procedures were used to assess S-T structure and its relation to direction selectivity. In the first, the S-T orientations of receptive fields were quantified by fitting response temporal phase versus stimulus spatial phase data. In the second procedure, conventional linear predictions of direction selectivity were computed from the amplitudes and phases of responses to stationary gratings. Extracellular recording locations were reconstructed histologically. Among direction-selective cells, S-T orientation was greatest in layer 4B and it correlated well (r = 0.76) with direction selectivity. In layer 6, S-T orientation was uniformly low, overlapping little with layer 4B, and it was not correlated with directional tuning. Layer 4A was intermediate in S-T orientation and its relation (r = 0.46) to direction selectivity. The same laminar patterns were observed using conventional linear predictions. The patterns do not reflect laminar differences in direction selectivity since the layers were equivalent in directional tuning. We also evaluated a model of linear spatiotemporal summation followed by a static nonlinear amplification (exponent model) to account for direction selectivity. The values of the exponents were estimated from differences between linearly predicted and actual amplitude modulations to counterphasing gratings. Comparing these exponents with another exponent—that required to obtain perfect matches between linearly predicted and measured directional tuning—indicates that an exponent model largely accounts for direction selectivity in most cells in layer 4, particularly layer 4B, but not in layer 6. Dynamic nonlinearities seem essential for cells in layer 6. We suggest that these laminar differences may partly reflect the differential involvement of geniculocortical and intracortical mechanisms.

Onset and offset of calcium current in response to short, depolarizing voltage pulses were studied in giant synaptic terminals of goldfish bipolar neurons. Tail-current decays were extremely rapid (80 μs at −70 mV), and exponentially slowed at more positive repolarization potentials. The amplitude of tail current following voltage pulses varied with the pulse amplitude, according to a Bolzmann distribution with a V1/2 of −27.6 mV and a slope factor of 13.8 mV. When interpreted with a Hodgkin-Huxley model, calcium-current onset was best described by m2 kinetics and a time constant (τm) of 0.62 ms at −10 mV. The kinetics of calcium-current onset and offset are at least two- to threefold faster than those described in other native cells. Combined with other recent data, the results suggest that the rapid kinetics may participate in a fast signaling mode in the goldfish ON-center pathway.

To assess the dolphin's capacity for color vision and determine the absorption maxima of the dolphin visual pigments, we have cloned and expressed the dolphin opsin genes. On the basis of sequence homology with other mammalian opsins, a dolphin rod and long-wavelength sensitive (LWS) cone opsin cDNAs were identified. Both dolphin opsin cDNAs were expressed in mammalian COS-7 cells. The resulting proteins were reconstituted with the chromophore 11-cis-retinal resulting in functional pigments with absorption maxima (λmax) of 488 and 524 nm for the rod and cone pigments respectively. These λmax values are considerably blue shifted compared to those of many terrestrial mammals. Although the dolphin possesses a gene homologous to other mammalian short-wavelength sensitive (SWS) opsins, it is not expressed in vivo and has accumulated a number of deletions, including a frame-shift mutation at nucleotide position 31. The dolphin therefore lacks the common dichromatic form of color vision typical of most terrestrial mammals.

We studied the interaction between the chromatic and temporal properties of parvocellular (P) neurons in the lateral geniculate nucleus (LGN) of macaque monkeys. We measured the amplitudes and phases of responses to stimulation by spatially uniform fields modulated sinusoidally about a white point in a three-dimensional color space, at a range of temporal frequencies between 1 and 25 Hz. Below about 4 Hz, temporal frequency had relatively little effect on chromatic tuning. At higher frequencies chromatic opponency was weakened in almost all cells. The complex interactions between temporal and chromatic properties are represented by a linear filter model that describes response amplitude and phase as a function of temporal frequency and direction in color space along which stimuli are modulated. The model stipulates the cone inputs to center and surround, their temporal properties, and the linear combination of center and surround signals. It predicts the amplitudes and phases of responses of P-cells, and the change of chromatic properties with temporal frequency. We used the model to investigate whether or not the chromatic signature of the surround in a red–green cell could be estimated from the change in the cell's chromatic properties with temporal frequency. Our findings could be equally well described by mixed cone surrounds as by pure cone surrounds, and we conclude that, with regard to temporal properties, there is no benefit to be gained by segregating cone classes in center and surround.

The metabotropic receptor mGluR6 is localized to the dendrites of On bipolar cells and mediates synaptic input from photoreceptors. The binding of glutamate to the receptor activates a phosphodiesterase (PDE), which then hydrolyzes cGMP. A nonselective cationic conductance, believed to be gated directly by cGMP, is turned off as a result of the fall in cGMP levels, and the cell hyperpolarizes. Here we present evidence for regulation of the conductance by an additional mechanism that it is independent of cGMP. Whole-cell recordings were obtained from On bipolar cells in slices of tiger salamander retina. Dialysis of cells with 1 μM KN-62 or 10 μM KN-93, two inhibitors of type II calmodulin-dependent protein kinase (CaMKII), depressed cGMP-dependent currents. This depression persisted when hydrolysis of cGMP was prevented with IBMX, a broad-spectrum PDE inhibitor, suggesting that CaMKII acts downstream from the PDE in the cascade. The depression of cGMP-dependent currents was probably not due to a direct interaction of the inhibitors with the channels as neither 1 μM KN-62 or 10 μM KN-93 was found to have any effect on cyclic nucleotide-gated channels when applied directly to excised patches of rod outer segments. We propose that phosphorylation by CaMKII may be an important mechanism for regulating the cGMP-dependent conductance of On bipolar cells.

Controversy over the relationship between ocular pigmentation and absolute dark-adapted light sensitivity has persisted for over two decades. Previous electrophysiological experiments in hypopigmented mammals (mice, rats, rabbits) show increased thresholds in the dark-adapted state proportional to the deficit in ocular melanin. Animals with the least amount of ocular melanin have the most elevated thresholds. Dark-adapted thresholds in hypopigmented mice show similar threshold elevations in behavioral tests. The present study extends these findings to show that a specific increase in ocular pigmentation results in the converse effect, lowered absolute dark-adapted thresholds. The increase in ocular melanin was accomplished by keeping Himalayan mice in the cold (4°C) for 6 weeks. Himalayan mice (C57BL/6J cH/cH) were compared to black mice (C57BL/6J +/+) and albino mice (C57BL/6J c2J/c2J) after 6 weeks at either 4°C or 20°C in 12-h cycling light (<1 cd/m2). The Himalayan mice that were kept in the cold exhibited a 44% increase in ocular melanin compared to Himalayan mice kept at room temperature. Cold rearing did not effect ocular melanin or visual thresholds in control animals (black mice = 10−5.9 cd/m2 and albino mice = 10−4.4 cd/m2). In contrast, the Himalayan mice maintained at 4°C had thresholds of 10−5.7 cd/m2 compared to 10−5.1 cd/m2 for Himalayan mice kept at 20°C. This represents compelling evidence of a direct relationship between ocular melanin concentration and absolute dark-adapted light sensitivity.

Müller cells are highly permeable to potassium ions and play a major role in maintaining potassium homeostasis in the vertebrate retina during light-evoked neuronal activity. Potassium fluxes across the Müller cell's membrane are believed to underlie the light-evoked responses of these cells. We studied the potassium currents of turtle Müller cells in the retinal slice and in dissociated cell preparations and their role in the genesis of the light-evoked responses of these cells. In either preparation, the I–V curve, measured under voltage-clamp conditions, consisted of inward and outward currents. A mixture of cesium ions, TEA, and 4-AP blocked the inward current but had no effect on the outward current. Extracellular cesium ions alone blocked the inward current but exerted no effect on the photoresponses. Extracellular barium ions blocked both inward and outward currents, induced substantial depolarization, and augmented the light-evoked responses, especially the OFF component. Exposing isolated Müller cells to a high potassium concentration did not cause any current or voltage responses when barium ions were present. In contrast, application of glutamate in the presence of barium ions induced a small inward current that was associated with a substantially augmented depolarizing wave relative to that observed under control conditions. This observation suggests a role for an electrogenic glutamate transporter in generating the OFF component of the turtle Müller cell photoresponse.

The inhibition of [3H]acetylcholine (ACh) release from cholinergic amacrine cells by glycine and GABA was studied using an in vivo eyecup preparation in anesthetized rabbits. Glycine (1 mM) had no effect on basal ACh release, but completely blocked the light-evoked release of ACh. Glycine also blocked the strong potentiating effects of picrotoxin (20 μM) normally observed on basal and light-evoked release. Strychnine (20 μM) increased basal release, albeit less than picrotoxin, but partially inhibited and altered the shape of light-evoked responses. Co-perfusion of picrotoxin and strychnine after strychnine application resulted in a larger additional basal increase. However, light-evoked responses were not restored to a control shape and magnitude, or to potentiated levels as with picrotoxin alone, but remained altered and partially inhibited. These results support the concept of a sustained GABA-mediated inhibition of the cholinergic pathway in the intact retina. In contrast, glycine-mediated inhibition of the cholinergic pathway differs, with the present results indicating a significantly smaller sustained inhibition of basal release and a temporal inhibition of light-evoked release. The lack of effect of any of these compounds on kainate-evoked responses indicates that these effects are predominately indirect, possibly on the presynaptic bipolar cell.

Photopic electroretinograms (ERGs) elicited by light flashes were recorded for three normal human subjects who were exposed, throughout, to natural ambient light cycles over 24-h day–night periods. ERGs were recorded either with the adaptation state of the eyes maintained at the level set by the natural ambient lighting, or after 10 min dark adaptation. The amplitudes and implicit times of both the a- and b-wave components of the ERG were examined and of these, only the b-wave implicit time exhibited significant diurnal variation, such that the nighttime values were 20–40% greater than those recorded during daytime. Such diurnal variations were observed under both recording conditions and cannot, therefore, be attributed to diurnal changes in the adaptation state of the cone photoreceptors. ERGs were recorded at midday and midnight during 24-h exposure to the natural light cycle, but during the recording period, the short-term adaptation state of the eye was controlled by exposure to rod saturating background field, so that visual sensitivity was the same at both recording times. The b-wave implicit times recorded at midnight were, nonetheless, greater than those recorded at midday. This difference is not, therefore, determined by the short-term state of retinal adaptation, but reflects long-term light history. Measurements performed under 24-h continuous light exposure showed no variation in the b-wave implicit time, whereas some measurements made during extended dark adaptation provided limited evidence for implicit time changes. By controlling the wavelengths to which the eye was exposed during the daylight phase of the diurnal cycle, it was shown that the shifts in b-wave implicit time associated with the change from dark to light are triggered by the rod system, although they are most clearly observed in the cone-dominant responses to long-wavelength light. The results demonstrate a diurnal variation in the temporal responses of the post-photoreceptoral cone pathways of the human retina, which is triggered by activation of the rod photoreceptors.

Reverse correlation was used in conjunction with ternary white noise to estimate the first-order spatiotemporal receptive-field structure of LGN cells in the anesthetized, paralyzed cat. Based on a singular-value decomposition of these data, we conclude that most LGN cells are approximately space–time separable. An analysis of the timecourses of the first singular values revealed a strongly bimodal but continuous distribution of rise times and waveforms. The two modes represented cells generally associated with the lagged and nonlagged classes of Mastronarde (1987a,b), and this was confirmed by their responses to step and sine-modulated spots in their field centers. The intermediate cells, rather than appearing to constitute a separate group, smoothly filled the region between the obviously lagged and nonlagged cells in every respect. These conclusions are limited to X-cells although the data from a much smaller population of Y-cells conform to the same scheme. We conclude that lagged and nonlagged cells represent the modes of a continuous and very broad distribution of temporal responses in the cat LGN.

Electrophysiological studies of the isolated retina involve perfusing the tissue with a physiological Ringer's. Organic pH buffers such as HEPES have become increasingly popular in recent years because for many purposes they offer a convenient and reliable alternative to the more traditional bicarbonate/CO2. In this paper, however, we report that important functional properties of rods, bipolar cells, and horizontal cells in the salamander, Ambystoma tigrinum, are sensitive to the choice of buffer and, in the case of horizontal cells, that sensitivity is acute. In bicarbonate/CO2 Ringer's, the dark potential of the horizontal cell was typically near −50 mV and saturating light caused it to hyperpolarize to about −75 mV. On switching to HEPES-buffered Ringer's at the same pH, horizontal cells depolarized in darkness to about −20 mV, close to the chloride equilibrium potential, and the kinetics of their light responses changed. The cone-driven components of light responses increased in size relative to rod-driven components. Saturating lights still hyperpolarized the cells to −75 mV, however. Horizontal cells, being coupled via gap junctions, form a syncytium and syncytial length constants, measured in bicarbonate/CO2 Ringer's, were generally in the range 150–225 μm. On switching to HEPES-buffered Ringer's, length constants increased substantially to 250–330 μm. All these changes were reversible. We discuss our findings within the context of the cell's ability to regulate its internal pH.

Recovery kinetics of the saturating photocurrent response in amphibian rods suggest regulation of the visual signal by a first-order deactivation reaction with an exponential time constant (τc) of about 2 s. The original hypothesis that τc represents the lifetime of activated rhodopsin (R*) in a single-step deactivation appears at odds with several recent findings, for example, that Ca2+, a known regulator of the enzymatic phosphorylation of R*, does not regulate the value of τc. A recently proposed alternative hypothesis, that τc is the lifetime of activated transducin and that the R* lifetime is relatively short (∼0.4 s), appears consistent with the Ca2+ data but is difficult to reconcile with a high specific catalytic activity of R*. The present theoretical study proposes a rate-equation model of R* activation and deactivation in amphibian rods that is generally consistent with observed properties of the τc-associated reaction and the action of Ca2+ as well as with the stereotyped nature of the single-photon response. The model is developed by considering the effect of background light on a time-dependent variable, Reff*, defined as the effective total level of R* activity. Central starting assumptions are that Ca2+ reduction mediates the effect of background light on Reff*(t) and that background desensitization of the photocurrent flash response derives from this action of Ca2+. Construction of the model is guided by criteria based on previous experimental findings. Among these are the approximate constancy of background desensitization expressed at near-peak and later times in the flash response, and the large (∼10-fold) dynamic range of this desensitization. The proposed model hypothesizes that an event regulated by Ca2+ feedback causes activated rhodopsin to become susceptible to a two-phase, stochastic deactivation process, the second phase of which is characterized by τc. A central prediction of the model is the regulated transition of flash-activated R* to “R**”, a state exhibiting greatly increased catalytic activity.

The ascending projections of the optic tectum, including their cells of origin, have been studied in the lizard Podarcis hispanica by means of a two-step experimental procedure. First, tracers were injected in the tectum to study the anterograde labeling in the forebrain. Second, the cells of origin of these projections have been identified by analyzing the retrograde labeling after tracer injections in the thalamus, hypothalamus, and pretectum. Three main tectal ascending pathways have been described: the dorsal tecto-thalamic tract (dtt), the medial tecto-thalamic tract (mtt), and the ventral tecto-thalamic tract (vtt). The dtt originates in radial cells of layers 5 and 7 and bipolar cells of layers 8 and 10 that project to the lateral neuropile of the dorsal lateral geniculate nucleus (GLD), to the intergeniculate leaflet (IGL), and to the ventral lateral geniculate nucleus (GLV). The mtt arises from radial neurons of layers 3 and 5 and bilaterally reaches the putative reticular thalamus and its boundary with the hypothalamus, the rostral IGL, and the area triangularis (AT). The vtt is composed of fibers from ganglion and multipolar cells of the layer 7 that project bilaterally to the nucleus of the vtt, the ventrolateral thalamic nucleus, the medial posterior thalamic nucleus (MP), the nucleus rotundus (Rot), the IGL, and the cell plate of the GLD. Therefore, the GLD receives not only direct retinal afferents but also two different tectal inputs, thus constituting a convergence point in the two visual pathways to the telencephalon. Moreover, different tectal cells specifically project to the ventrolateral thalamus and to pretectal nuclei. These results are discussed from comparative and functional viewpoints.

Patch-clamp and calcium-indicator measurements were used to examine the electrical excitability of large-terminal bipolar neurons from goldfish retina. Large, transient increases in intracellular calcium occurred spontaneously in the synaptic terminal but not in the soma of bipolar neurons. Calcium transients were blocked by hyperpolarization, by external application of calcium-channel blockers, and by the neurotransmitters GABA and glutamate. These observations suggest that calcium action potentials are responsible for the spontaneous increases in intraterminal calcium, which was directly confirmed by electrical recordings of calcium-dependent action potentials in both whole-cell and perforated-patch recordings. We suggest that regenerative depolarization produced by the opening of calcium channels in the synaptic terminal of on-type bipolar neurons represents an amplification mechanism in the high-sensitivity ON pathway in the dark-adapted fish retina.
