Clastogenic Effect of TrenimonR on in Vitro Chromosomes of Phenotypically Healthy Subjects with Morphologically Abnormal Caryotype*

Abstract
The effect of a given concentration of TrenimonR (6 × 20−8 mol/1) on the lymphocytes of 11 healthy subjects with morphologically abnormal karyotype was analysed in vitro. The research included 6 subjects with balanced translocations, one of them with a centric fusion. Of the remaining 5 subjects, 2 had a duplication in the paracentric region, 1 showed an inversion, and 2 had a marker chromosome.
In the group with morphologically abnormal chromosomes a higher sensibility to the influence of TrenimonR was found as compared with a control group with morphologically normal karyotype. The subjects with an atypical karyotype had 36 to 65% aberrations, the control group 28%. A localization of the breaks showed no increased aberration rates in the morphologically abnormal chromosomes.
Zusammenfassung
Der Effekt einer ausgewählten Konzentration von TrenimonR (6 × 10−8 Mol/1) auf die Lymphozyten von 1 gesunden Personen mit morphologisch abnormem Karyotyp wurde in vitro untersucht. Einbezogen ware sechs Personen mit balancierten Translokationen, davon eine zentrische Fusion, zwei Personen mit Verdoppelung des centromernahen Abschnittes, eine Person mit einer Inversion und zwei Träger von Markerchromosomen. Es fand sich eine erhöhte Sensibilität der Gruppe mit morphologischen Chromosomenveränderungen gegenüber der Einwirkung von Trenimon im Vergleich zu einer Kontrollgruppe mit morphologisch unauffälligem Karyotyp. Der Prozentsatz der Zellen mit Aberrationen lag dabei bei den Probanden mit atypischem Karyogramm zwischen 36 und 65%, bei den Kontrollpersonen zwischen 28 und 58%. Die gleichfalls durchgeführte Bruchlokalisation zeigte keine erhöhte Beteiligung der morphologisch veränderten Chromosomen an den Aberrationsraten.
Riassunto
È stato studiato l'effetto in vitro di una data concentrazione di TrenimonR (6 × 10−8 mol/1) sui linfociti di 11 soggetti con cariotipo morfologicamente anormale. Si è osservata una traslocazione bilanciata in 6 soggetti (di cui uno con fusione centrica), una duplicazione della regione paracentrica in 2 soggetti, un'inversione in 1 soggetto, ed un cromosoma marcatore in 2 soggetti. Si è inoltre osservato che il gruppo a cariotipo morfologicamente anormale si dimostrava piú sensibile all'azione del Trenimon rispetto al gruppo di controllo. Nei soggetti a cariotipo anormale la percentuale di cellule con aberrazioni oscillava tra il 36 e il 65%, contro il 28-58% dei soggetti di controllo. La contemporanea localizzazione di rotture non presentava alcun aumento del tasso di aberrazione nei cromosomi morfologicamente anormali.
Résumé
L'effet d'une concentration déterminée de TrenimonR (6 × 10−8 mol/1) dans les lymphocytes de 11 sujets sains avec karyotype morphologiquement anormal fut analysé “in vitro”. On observa une translocation balancée chez 6 sujets (dont un comprenant une fusion centrique), une duplication de la région paracentrique chez 2 sujets, une inversion chez 1 sujet, et un chromosome marqueur chez 2 sujets. On observa de plus que le groupe à karyotype morphologiquement anormal montrait une plus grande sensibilité à l'action du Trenimon que le groupe de contrôle à karyotype morphologiquement normal. Chez les sujets à karyotype anormal, le pourcentage de cellules avec aberrations oscillait entre 36 et 65 %, et entre 28 et 58 % chez les sujets de contrôle. La localisation contemporaine de ruptures ne montra aucune augmentation des taux d'aberration dans les chromosomes morphologiquement anormaux.
